Part J119335 but replaced the M6" riboswitch with the newly designed M6"-R2 riboswitch.
This construct is a basic biosensor for ammeline, based on part J119335. The P5 promoter drives the M6"-R3 riboswitch, which permits translation of the reporter gene (RFP) in the presence of ammeline.
The M6"-R3 riboswitch is based on the M6" riboswitch, with a strong RBS (AGGAGAA) and sequence that is designed to completely sequester the Shine-Dalgarno sequence when ammeline is absent (see Figure 1A of http://www.pnas.org/content/107/7/2830.full).
Mutation at position 430 nt of A to G changes tyrosine (Y) to cysteine (C) in RFP CDS.
