The operon used comes from Xenorhabdus nematophila, a nematode-dependent parasite. This operon allows the formation of a biofilm on nematodes pathogenic to insects and it is thus key in this nematode-bacteria symbiotic association that kills insects.
To control the expression of the operon, we used a promoter from the standard registry of parts, which was characterized by our team last year (see Part:BBa_K763003).
The molecular mechanism works as follows: with low nitrogen in the media, the promoter is activated and a biofilm of E.coli over C.elegans should be triggered and with high nitrogen concentration, the biobrick is unactive and Pseudomonas will not form the biofilm.

We developed a nitrogen-gradation tube experiment, which means that we added different (NH4)2SO4 concentrations to each tube, so we can measure a gradation of the protein fluorescence. We used a synthetic medium for this.
