The sequence was obtained by applying all mutations that upregulated AraC binding and subsequent promoter activity listed in reference [1].

pBAD is an <i>E.coli</i> promoter that is induced by L-arabinose. K206000 is a mutagenized pBAD promoter that is responsive to lower concentrations of arabinose than wild type (<partinfo>I13453</partinfo>) and, additionally, exhibits a higher maximum of protein expression as measured by coupling to a fluorescent reporter.

KZ144 Endolysin from Pseudomonas aeruginosa bacteriophage
2013-09-15T11:00:00Z
2015-05-08T01:09:21Z
This part comes from genomic DNA sequence of Pseudomonas aeruginosa bacteriophage, ΦKZ.
This part is the sequence for the protein made from a bacteriophage that targets Pseudomonas aeruginosa and is responsible for lysing the bacteria. This protein is one of many kinds of endolysins. Endolysins typically have a high specificity and bacteria have not generally shown to become resistant to these proteins. There have been studies on how to vary the activity and specificity of this endolysin in particular, making it lyse other gram negative pathogens. This can be used in future projects that require the lysing of bacterial cells.

An L-Arabinose controlled endolysin from bacteriophage ΦKZ
2013-09-15T11:00:00Z
2015-05-08T01:09:21Z
The endolysin gene is from bacteriophage ΦKZ. An E. coli promoter and terminator were used.
L-arabinose induced KZ144 endolysin is a functional construct which contains the gene that encodes for the endolysin protein from the bacteriophage ΦKZ (BBa_K1136002) which targets Pseudomonas aeruginosa. The construct is under the control of an L-arabinose promoter (BBa_K206000) and the rrnb terminator (BBa_B0010). This part can be expressed in E. coli using 20% L-Arabinose.

The endolysin gene was synthesized by GenScript to optimize codons and avoid illegal sites. The promotor and terminator were chosen for controllable expression, because the endolysin may be toxic in high copy number.
