<P> <P>Suggested by Sri Kosuri and Drew Endy as a high efficiency terminator. The 5' end cutoff was placed immediately after the TAA stop codon and the 3' end cutoff was placed just prior to the RBS of Gene 1.4 (before AAGGAG).<P> Use anywhere transcription should be stopped when the gene of interest is upstream of this terminator.

The GFP present in this part is modified from (BBa_K208000 http://parts.igem.org/wiki/index.php?title=Part:BBa_K208000), with stop codon removed to allow for full translation to 10x His Tag, BBa_K844000 (http://parts.igem.org/wiki/index.php?title=Part:BBa_K844000). This part is useful as it allows the user to carry out gene expression studies and purify the final protein-GFP-His Tag fusion if desired. The prefix in front of the GFP is RFC 23 compatible to allow for in-frame expression of genes.
Once protein expression has been demonstrated the gene can be put in front of BBa_K844000 for protein purification without the GFP.
