This part was extracted from [[Part:pSB2K3|pSB2K3]] and is described on NCBI at accession number YP_006518.
The P1 lytic replication region, from Enterobacteria phage P1, contains RepL which initiates replication at origin L (interior to RepL). It is regulated by a lacI promoter. When present in a cell producing LacI, the copy number is <10 copies per cell. If IPTG is added to the media, the promoter is "un-locked", causing the plasmid to be produced at >100+ copies per cell.
