Users should check for secondary structures induced in the RBS by upstream and downstream elements in the +50 to -50 region, as such structures will greatly affect the strength of the RBS.

I-F type CRISPR-Cas system was cloned from Aggregatibacter actinomycetemcomitans D7-S strand.
Gene encoding I-F type CRISPR_Cas system Cas3 protein, which is an helicase and ssDNA nuclease. Attached there is a pLux/cI right promoter and strong RBS site. In the I-F type CRISPR-Cas system Cascade complex distinguishes target DNA, and rectruits Cas3 protein, which acts as a nuclease and helicase, which hidrolises ssDNA in a 3'-5' direction.
