rtTA is a regulatory protein which can combined with tetracycline analogues. Like the protein in Biological signaling pathways. When it combine with tetracycline analogues, it can change its conformation to activate the subsequent signal path. Under the action of constitutive promoter, the regulatory protein expresses stably. When it combine with tetracycline analogues, rtTA change its conformation to activate the promoter, then the report gene expresses.
