The sequence of this part was confirmed by Sanger Sequencing. RBS* is a ribosome binding site that has been specifically designed to include the anti-Shine Dalgarno (SD) sequence of Synechocystis sp. strain PCC 6803. If no extra bases are added to its sequence, RBS* has an optimal spacing of 9bp between the central base of the core SD sequence (which is base A at position 7 of its sequence) and the first base of the start codon (1). RBS* has been shown to be 5 times stronger than BBa_B0034 in Synechocystis (1), making it the strongest currently available RBS for heterologous gene expression in this cyanobacterium.
