Homo sapiens adrenoceptor beta 2 (ADRB2) is a G-protein coupled receptor (GPCR) activated by adrenaline that plays an important part in cardiovascular and pulmonary physiology, and is one of the most extensively characterized members of this large family of membrane proteins. It has seven membrane-spanning helices with an extracellular N-terminus and an intracellular C-terminus like all other members from GPCR family. In its host cell, once stimulated by catecholamine (epinephrine/norepinephrine), ADRB2 will undergo conformational change and interact with heterotrimeric G proteins to make alpha subunit (Gs) disassociated, then Gs will activate the adenylyl cyclase and further result in the soaring intracellular cAMP level and activation of enzymes downstream. It is reported to be functionally expressed in many mammalian and insect cells.

First, it has no obvious tag for researchers to detect it directly, so we further add a 6xhis tag at its C terminus. Second, as the N terminus sequence may decide the membrane localization in different host cells, we used the C terminus sequence of GPCR RI7 and STE2 which were both reported to be successfully located to yeast membrane to replace the corresponding sequence of ADRB2.
