It is ordered via IDT sponsorship. Anti-meth scFv is coming from mus musculus. YFP and CFP are the common fluorescent proteins which are derivatives of GFP.
This construct consists one domain of anti-meth scFv antibody (termed as c1 in the project) conjugated with YFP, and the other domain of anti-meth scFv antibody (termed as c2 in the project) conjugated with CFP. Also, the construct has its own T7 promoter sequence in addition to the T7 promoter in the vector it is cloned into. This design constructed with the thought of separately producing the two domains of the anti-meth antibody which are conjugated with fluorescence proteins YFP and CFP, when the antibody domains came together in the presence of methamphetamine, it will give a FRET (f??rster resonance energy transfer) emission between CFP and YFP.
