Cactag - like a biobrick scar but first T changed to C.
2010-08-10T11:00:00Z
2015-05-08T01:11:50Z
Use only if the next part starts with ATG. This is a virtual part useful to anyone working with Anderson's RBS library.
First nucleotide of the biobrick suffix is varies in the library. Some RBSes have T, wchich results in a regular suffix, but other hace c,G or A. This modified scar is to help in sillico assembly of the constructs that use Anderson's RBSes.
This modified scar is suitable only if the next part starts with ATG

Suggested by Sri Kosuri and Drew Endy as a high efficiency terminator. The 5' end cutoff was placed immediately after the TAA stop codon and the 3' end cutoff was placed just prior to the RBS of Gene 1.4 (before AAGGAG).
Use anywhere transcription should be stopped when the gene of interest is upstream of this terminator.

RBS measurement device pT7 J61107 GFP
2010-10-15T11:00:00Z
2015-05-08T01:11:50Z
This part contains RBS J61107 from the Anderson's collection.
In the physical DNA after J61107 in composite parts there is Cactag sequence instead of the biobrick scar Tactag. J61101 was originally designed in a way that its last nucleotide "C" is also first nucleotide of the suffix (wchich should be T)- it modifies the suffix. The sequence of J61101 in the registry does not include its last nucleotide. If it were included the registry sequence would show the additional nucleotide, now it shows T instead of C.
The seguence of the J612101 does not match its physical DNA
To go around this problem in out bricks we used scarless assembly option and manually included the scars:
BBa_K299800 DNA standard biobrick scar (if next part starts with ATG)
BBa_K299801 DNA standard biobrick scar (if next sequence does NOT start with ATG)
BBa_K299802 DNA Aactag - like a biobrick scar but first T changed to A
BBa_K299803 DNA Gactag - like a biobrick scar but first T changed to G
BBa_K299804 DNA Cactag - like a biobrick scar but first T changed to C.
This brick consists of the following subparts:
I719005[T7promoter] BBa_K299801[standard scar] BBa_J61107[RBS] BBa_K299804[Cactag] BBa_E0040[GFP] BBa_K299801[standard scar] BBa_B0010[terminator] BBa_K299801[standard scar] BBa_B0012[terminator]
