The backbone of our part BBa-K341000_eGFP_recombination is PSB1C13, resistant to Amp. Restriction enzyme I-SceI cutting site, Recombination sequence 1, homologous to part of modified E.coli genome, coding sequence of gene eGFP, recombination sequence 2, homologous to another part of modified E.coli genome, I-Scel cutting site( the elements detailed above are listed in the order from 5??? to 3???) are added into the backbone with EcoR I and Pst I. The biobrick we construct in this part is to verify landing pad method, on which our project is based on.
