This part was made with the 35S from BBa_K414002 (Wheeler, 2010) who obtained it from pK7FWG2 plasmid. Also the GFP with strong plant RBS was taken from BBa_K414001 (Bersaba, 2010).
This composite was designed to bring together a constitutive plant promoter (35S) with a fluorescent reporter (GFP) that has a strong plant RBS (Kozak)upstream to maximize translational efficiency. A composite part like this one may be used as an internal control in plant transformations to measure relative, induced expression of an inducible gene.

The Kozak sequence was directly engineered into the prefix primer and used to PCR out the GFP gene from E0040.
In order for Green Fluorescent Protein to function properly in plants, a plant-specific ribosome binding site (Kozak sequence)must be added to the 5'region just upstream from the start of translation.The Kozak sequence is AAA AAA AAA ACA and is directly adjacent to the ATG start codon in GFP.
