The LacI repressor was PCR amplified from a plasmid source of a biobrick (BBa_K319046) that was submitted last year by the uOttawa team. Yeast codon optimized BFP was synthesized by the uOttawa team using Mr. Gene DNA synthesis. The rtTA activation domain was PCR amplified from a plasmid obtained by our host lab.
LacI repressor tagged upstream with yeast codon optimized blue fluorescent protein and an rtTA activation domain.
