This is a standard sequence, very used in many constructs.
As i don't have any design considerations i can say that generally to make screening gels of such short segments is preferable to use TBE1X instead of TAE1X.

The Ptac promoter has been inserted with a PCR, the sequence of the promoter has been taken from literature,
while the original plamid is pET21b
This is the twin construct of BBa_K731700, with the same two subsequent fluorescent proteins, with T7 promoter replaced with Ptac. This construct allows to test possible differences in terminator efficiency due to different promoter and thus polymerase.
We tested both constructs in the E.Coli strain BL21pLysS, that has both polymerases.

The presence of the promoter cannot be tested with screening digestion, as it has more or less the same T7 promoter`s length. Anyway the screening in amplification strains is much more easy: the right colonies will be intensely red.
