Our research has focused on novel biosynthetic pathways for UV-protectant compounds found in two distinct algal species. One leading to the production of the unrelated UV-screening compound scytonemin was cloned from Nostoc punctiforme. ScyB first converts tryptophan into 3-indole pyruvic acid, which is converted into a ketoacid intermediate by ScyA. ScyC then converts the ketoacid into an indole compound which spontaneously forms scytonemin.
This part is an intermediate generated during the making of BBa_K814011. It contains the scyB and scyC generators, but because it lacks scyA, it is not expected to produce scytonemin.

N. punctiforme genomic DNA was obtained from ATCC. From here, PCR primer extension was used to clone each gene with flanking restriction digest sites to be used for vector cloning. The genes were cloned into an individual pUCBB BioBrick??? cloning vectors containing a constitutive modified lac promoter (lacP???) and an RBS. The resulting BioBrick??? parts were then assembled using the standard assembly protocol.
