Our research has focused onnovel biosynthetic pathways for UV-protectant compounds found in two distinct algal species. One leading to the production of the unrelated UV-screening compound scytonemin was cloned from Nostoc punctiforme. ScyC (NpR1276) is a protein of unknown function which converts 3-indole pyruvic acid to a ketoacid intermediate which spontaneously dimerizes to form scytonemin.
This part includes a modified constitutive lac promoter (lacP'), and RBS and the open reading frame of scyC. This part can be used to express ScyC protein in E. coli.

N. punctiforme genomic DNA was obtained from ATCC. From here, PCR primer extension was used to clone the gene with flanking restriction digest sites to be used for vector cloning. The gene was cloned into an individual pUCBB BioBrick??? cloning vector containing a constitutive modified lac promoter (lacP???) and an RBS.

Our research has focused onnovel biosynthetic pathways for UV-protectant compounds found in two distinct algal species. One leading to the production of the unrelated UV-screening compound scytonemin was cloned from Nostoc punctiforme. ScyB first converts tryptophan into 3-indole pyruvic acid, which is converted into a ketoacid intermediate by ScyA. ScyC then converts the ketoacid into an indole compound which spontaneously forms scytonemin.
This part is made of three basic parts and contains the scyB, scyA and scyC generators. This part could be used for scytonemin biosynthesis in E. coli
