This part is a component of the vector included with the Invitrogen pBAD/Thio-TOPO kit.
This is a tag which is added to the C terminus of a protein in order to aid in the identification of the protein in vivo, as well as to simplify the purificaiton process of the protein. The V5 epitope bind the Anti-V5 antibody. The histidine tag allows for purification using a nickel column. It was sourced from the Invitrogen pBAD/Thio-TOPO, which enables the expression of PCR amplified DNA. This protein also includes a linker to attach the V5 and histidine regions with the protein that is being expressed.

ZCD originated from Crocus Sativus, the saffron plant. However the version present here has been codon optimized for synechocystis.
This is the functional unit which makes ZCD soluble in E. coli. It includes a inducible promoter, mutated thioredoxin to increase solubility, the ZCD ORF, a hist-tag for easy purification and finally a double terminator. This part has been shown to highly express ZCD in E. coli. It is suggested that this part be used in a plasmid also containing araC for the optimal repression of this protein before the time of induction.

Zeaxanthin 7,8-cleavage dioxygenase (ZCD)

This enzyme was first identified in Crocus sativus, the saffron plant. The originally identified gene can be found here [http://www.ncbi.nlm.nih.gov/nuccore/AJ489276.1 AJ489276.1]
This is the enzyme required to cleave zeaxanthin at the 7, 8 position to begin the enzymatic process of making crocin, picrocrocin and safranal. ZCD seems to be unique to the species Crocus sativus, and has been found to be highly expressed in the style branch tissues of this species. Also, it has been shown to produce insoluble inclusion bodies when over expressed in E. coli. As a result, the functional expression of this protein requires the use of a soluble fusion proteins, such as thioredoxin, and induction at 20degrees.

This modified thioredoxin was produced by Zhijian Lu and collegues as described in the paper titled "Histidine Patch Thioredoxins

This is a mutant of E. coli's native thioredoxin. It has been modified to have the ability to chelate metal ions, notably nickel. This protein is useful as a fusion protein which can help to increase the solubility of difficult-to-express proteins and maximize yield of those proteins.
