NifHDK promoter follows the NifA transcription factor and comprises three structural genes nifK,nif D, and nifH. nifK encodes for B-subunit of Component 1 of nitrogenase. nifD encodes for alpha subunit of component 1 of nitrogenase. nifH encodes for component 2 of nitrogenase.

This sequence comes from the genome of Azotobacter vinelandii, specifically the nif cluster.
NifA protein is nitrogen senitive and regulates transcription of nif genes. When there isn't enough fixed nitrogen available for the organism's use, NifA is expressed, and NifA activates the rest of the nif genes. If there is a sufficient amount of reduced nitrogen or oxygen is present, another protein is activated: NifL. NifL inhibits the action of NifA and results in inhibition of nitrogenase formation.

The nifRLA promoter sequence was determined from the nif gene cluster sequence for Azotobacter vinelandii. It is found upstream of the nifL repressor.
The nifRLA operon regulates the expression of the nitrogen fixation (nif) genes. NifA activates transcription of nif genes by the alternative form of RNA-polymerase, s54-holoenzyme. NifL is a negative regulatory gene which inhibits the activation of other nif genes by nifA protein. NifR is a repressor binding site, between the promoter of the nifRLA operon and the nifL gene.
