Cop Y is a negative transcriptional regulatory protein. In the absence of effector molecules (in this case, CopZ and Cu2+), CopY binds to the promoter and depresses transcription of cop operon. On the other hand, CopZ is an anti-repressor of cop operon. Cop Z brings copper to CopY that is bound to the operon. In the presence of CopZ and copper, CopY detaches itself from the operon. When using CopZ, always pair it up with CopY, to get the copper sensing feature.

The whole CopYAZ operon from Streptococcus mutant allows the bacteria to sense and translocate copper in their system. CopY and CopZ are regulators of the expression of the cop operon in response to both copper starvation and copper excess, and CopA is a Cu(I)-translocating P-type ATPase. Since our project is to create a sensor, we concentrated on CopY and CopZ, which regulate transcription of cop operon.
