Description
The butanoate pathway in Clostridium acetobutylicum from acetyl-CoA to butanol requires five (or six) enzymes. In order, these are acetyl-CoA acetyltransferase (thiolase in E. coli), B-hydroxy butyryl coA dehydrogenase, Enoyl-coA hydratase (from E. coli), Butyryl coA Dehydrogenase, Butyraldehyde dehydrogenase, and Butanol dehydrogenase. It is unclear whether the E. coli thiolase will be sufficient for butanol production in E. coli or whether overexpression will be required. Subsequently, it is not included in this version of the operon.
Notes
We were unable to complete addition of the fifth gene to the operon (Butanol dehydrogenase). It needs to be cloned from I725025 to complete this.
