Description

The tCloneTet+PDB plasmid was obtained from part J100255 and was modified using Golden Gate Assembly to have a P2 promoter rather than the P5 promoter. The P2 promoter is known to have a lower efficiency than P5 but it is still relatively high compared to other promoters.

Notes

Used PCR to "cut out" the P5 promoter since there were no restriction enzyme sites surrounding it.
