Description

This vector is pSB1A2 with its BsaI site removed (BR = BsaI Removed), so it can be used in BsaI Golden Gate Assembly. It also has been verified to have a G at position -1 of the RNA I gene in the pMB1 origin of replication. This site has been identified as a hotspot for mutation of the pMB1 origin (Lin-Chao et al. 1992).
Note: Having a G in this position is considered to be the wild type sequence, and is the sequence listed in the Registry for pSB1A2.

Notes

Introduction of hotspot mutations was performed for the purpose of changing the plasmid copy number.
