will be made by adding an ligating a one sided insert into J70590 (that has been cut with BsaXI and PCR purified)

next cut with BsaXI and ligate again (will not need to purify)

problem will be if there is not enough left after first purification (unfortunately BsaXI cannot be heat killed)
