Description
In order to build up flagella usable as protein scaffold we wanted to design a system which makes it possible to switch a high variety of functional modules. For that purpose we designed a flagellin construct containing a Strep-Tag inserted into the D2 domain of Salmonella typhimurium on top of the Bacillus Subtilis flagellin as a linker region.
By exposure of the Strep-Tag to the outside of the flagellum it would be possible to connect Streptavidin-hybridized fusion proteins of various functionalities to the flagellum as scaffold increasing the epitope density.
