Description
NisT codes for a transporter in the membrane of Lactococcus lactis to transport nisin out of the cell. Together with the genes NisA, NisB, NisC, NisP, NisR and NisK it is responsible for producing the lantibiotic nisin in L. lactis.
Notes
Used primers:
forward - 5'-GTTTCTTCGAATTCGCGGCCGCTTCTAGATGGATGAAGTGAAAGAATTTACATCA-3'
reverse - 5'-GTTTCTTCCTGCAGCGGCCGCTACTAGTATTATTATTCATCATTATCCT-3'
to PCR NisT from the genome and add the BioBrick prefix and suffix.
Deleted illegal EcoRI site at position 44 using above primers, changing a TTC codon in a TTT. The illegal SpeI sites at position 980 and 1210 were removed using primers as well, and fusing the genes together again with a Gibson assembly.
Source
Obtained by PCR on the genome of Lactococcus lactis NZ9700, a nisin producing strain.
