Notes

The dinucleotide repeat was placed into synthesized primers that bound to the existing part I13401, and then a PCR reaction was run.

Source

With 22 AC repeats, the GFP sequence is -1 frame from the start codon, and so GFP should not be transcribed. The number of AC repeats was limited by maximation length of synthesized primers.
