Description
This part is a phagemid combined with RFP (BBa_J04450). Phagemid pBluescript is a special plasmid, it functions as a normal plasmid when transformed into bacteria. But when helper phage M13KO7 infect this bacteria, it will produce progeny M13 phage as vector for sequence on its MCS.
Phagemid pBluescript contains a strong package signal upstream on its MCS while M13KO7 helper phage contains a lower package signal for its genome. So after the infection of M13KO7, its genome can produce protein but these proteins will see pBluescript???s MCS as their true ???genome??? because pBluescript has a stronger package signal. As a result, the progeny phage???s genome will only contains pBluescript???s MCS, they cannot make next generation.
Here we put a reporter gene --- RFP (BBa_J04450) into our pBluescript, so when we transformed this phagemid into bacteria it should have red fluorescence. And if we add M13KO7 to infect this bacteria, other bacteria nearby should have red fluorescence, too.
