Description
This part is a phagemid. It functions as a normal plasmid when transformed into bacteria. But when helper phage M13KO7 infect this bacteria, it will produce progeny M13 phage.
Phagemid pBluescript contains a f1 ori while M13KO7 helper phage's package signal is modified as knock-down. So after the infection of M13KO7, its genome can produce protein but these proteins will see pBluescript as their true ???genome???. As a result, the progeny phage???s genome will only contains pBluescript, so they cannot produce next generation by infection.
We have removed some sequence on its MCS to create a linearized form of this phagemid.
Notes
Normally, a pBluescript contains EcoRI, PstI, XbaI and SpeI on its MCS. We have removed these sequence to create a linearized form of this phagemid.
Source
Its f1 ori comes from phage M13.
