Description

This construct can be used to knockout the acyl-CoA dehydrogenase 2 gene in Haloferax Volcanii through Thorston Allers' pop-in pop-out method(Halo Handbook). Thus the sequence is composed of 400 base pairs of homologous upstream genomic DNA, the first 50 base pairs of Acd2 in Haloferax Volcanii, 99 base pairs of nonsense DNA, the final 50 base pairs of Acd2, followed by 400 base pairs of homologous downstream genomic DNA. The product of successful plasmid integration and treatment with 5-FOA results in the Acd2 gene replaced by the 99 base pairs of nonsense DNA.
