Description
This aptameric sequence is capable of recognising and binding specifically to lead II ions. This sequence, which works as a single stranded nucleic acid molecule, is capable of forming a G-quadruplex by interacting with lead ions (Pb2+) while disrupting a partially complementary duplex.
Notes
For standardisation purposes and its usage in iGEM, this sequence was cloned as a duplex on pSB1C3 thereby adding the prefix and suffix sequences to it.
Given the fact that the sequence is need as a single-strand molecule, a primer was designed in order to carry out a PCR with a single primer for the production of this molecule.
Source
This sequence does not come from a genomic sequence. First reported by Li et al 2010, it is comprised by 16 nucleotides with a high percentage of G which are the ones capable of stabilising and forming the quadruplex with the lead ion.
