Description
LamB is a membrane channel that transports maltose and maltodextrins as well as the receptor for phage lambda. Metallothionein is a protein that sequesters 11 copper atoms and detoxifies radicals produced by copper. This part contains two genes fused together that allows for the expression of metallothionein on the outer surface of E. coli. CUP1, the gene coding for metallothionein from S. cerevisiae, is inserted into amino acid position 153 of the LamB gene sequence.
Notes
The stop codon of CUP1 was removed before being inserted into LamB. In making a G Block for this part to be synthesized, a 20 bp overlap with the plasmid pSB1C3 needed to be added to the ends of the sequence. This needed to be done to allow Gibson Assembly to yield our completed plasmid containing this part in pSB1C3.
