Description
This devise is consisted of a HSP70A-RBCS2 promoter, Cry10Aa toxin, extended FMDV 2A peptides, a Cyt1 toxin and also a terminator. The HSP70A-RBCS2 promoter is a heat-induced promoter. Both Cry and Cyt are pore-froming toxins from Bacillus thuringiensis produced as parasporal crystal components. The synergism between the two toxins was observed against dipteran insects such as mosquitoes. It has been proved that the synergism can contribute to the suppression of resistance. The FMDV 2A peptides serves as a linker between a upstream protein and a downstream protein in order to co-express them in a single vector. The device we designed are all codon-optimized so that we can safely express the toxins via Chlamydomonas reinhardtii.

Source
HSP70A-RBCS2 promoter cloned from Chlamydomonas reinhardtii. Cry10Aa and Cyt1 cloned from Bacillus thuringiensis serovar israelensis strain BRC-LLP29, codon optimal for Chlamydomonas reinhardtii. Extended FMDV 2A peptides cloned from FMDV, codon optimal for Chlamydomonas reinhardtii.
