Description
Isolation of the NPR reductase from the original plasmid caused differing restriction sites, bridged by oligonucleotides with common overhangs to both sites and a poly-A tail in between the overhangs. Subsequent ligation into the pSB1c3 plasmid gives the Biobrick preffix, followed by the NADPH Biobrick, followed by HindIII restriction site, and lastly the Biobrick suffix.
Notes
Oligo bridges between the restriction sites were noncoding and annealed solely for the purpose of standardizing the plasmid backbone.
