Description

This part is a gene circuit that can be used to produce the enzyme SmCPS1. SmCPS1 is a key enzyme on the biosynthesis pathway of tanshinone (a compound used to treat cardiovascular diseases). The tac promoter, a strong hybrid promoter, is used to control and increase the expression levels of the target gene (SmCPS1), and is used in the expression of recombinant proteins, produced from the combination of promoters from the trp and lac operons. The GST label is used to purify the recombinant protein. The GFP is used to measure the expression level of the part.

Notes

Two extra sticky ends are added to the gene when it is cloned via PCR. They are needed so that the part can be loaded onto other vectors. Site-directed mutagenesis is performed to alter the triplets identical to the sequence of restriction enzymes used to remove the part from the vector.

Source

SmCPS1 comes from the genome of Salvia miltiorrhiza.
