Description
This part is our "final" population counter with Tetracycline resistance, RFP fluorescence and timer device. However the cells express constitutively RFP without reasonable explanation...
When IntI1 (
BBa_K329014) is expressed, it excises the double terminator flanked by attC sites (
BBa_J99001) and triggers the expression of TetA(C) (with a medium RBS,
BBa_K329000), mRFP1 and LuxI (
BBa_K081008) because of the constitutive promoter upstream. LuxI produces HSL which binds to LuxR (expressed constitutively), then activates the transcription of GFP.
Notes
We used BioBrick Standard Assembly.
