Description
The gusA reporter gene encodes beta-glucuronidase (GUS), an enzyme from Escherichia coli.
Originally, the GUS reporter was designed to be used in plant biology to measure the activity of a promoter. The reporter was later adopted for cell screening and promoter measurement in bacteria. There are three often used methods: histochemical assay, fluorimetrical assay and spectrophotometric assay. The GUS substrates for these assays varies (5-bromo-4-chloro-3-indolyl glucuronide for histochemical assay, 4-methylumbelliferyl-beta-D-glucuronide for fluorimetrical assay, p-nitrophenyl β-D-glucuronide for spectrophotometric assay).
Notes
The part has been prefixed by EcoRI, XbaI and suffixed by SpeI, PstI according to the BioBrick??? standard.
Source
The gusA reporter gene comes from Escherichia coli uidA gene.
