Description
This is a fusion of muconolactone D-isomerase (Enzyme 3) and 3-oxoadipate enol-lactonase (Enzyme 4) in the catechol ortho-cleavage pathway. The proteins are fused c-terminal to n-terminal, respectively, with a rigid alpha-helical linker.
Notes
The selected linker is rigid to prevent the two fusion componenet enzymes from collapsing on each other. At the same time the fusion must display channeling capabilities.
Source
The part is constructed form the genomic sequence from P. Putida, optimized to E. Coli.
