Description

This is the reverse primer to PCR agrCA. Together with the forward primer (BBa_K391002), it is possible to PCR agrCA from S. aureus strain NCTC 8325. This primer adds SpeI and PstI sites to the PCR product. UBC iGEM was able to obtain correct bp PCR product, but was not able to clone the product to a BioBrick vector.

Notes

These primers flank naturally occuring RBS sites for agrCA. agrC is upstream of agrA in the genome. The PCR starts approximately 100 bp of agrC at a putative RBS site, ensuring that a RBS site is present. agrA, because of its location, will have a RBS upstream as well.
