Description
This origin is derived from the low (~20) copy pBR322 plasmid. The pMB1 replication origin for pBR322 is a regulated version of the high copy pUC19 origin (see I50022). The copy number is regulated by a small protein ROP. This part contains the origin with the appropriate copy-number regulatory domains, and can be used with the standard biobrick base vector I51020 to construct biobrick backbones. The part contains a RBS before the ROP protein but not a promotor.
Note: This part contains an VR sequencing site between the ROP and origin, due to its homology with the pUC19 origin. This site should ideally be removed in a revised version.
Notes
The extra reverse sequencing site in between the ROP and origin should be fixed in the future. Because of the sequence homologies with pUC19 origin backbones such as pSB1C3, it is impossible to specifically target the unwanted site with PCR without switching backbones.
