Description

This part encodes a transactivator composed of the DNA-binding domain for the tetracycline operator sequence and the viral VP16 transactivation domain; this protein has been engineered over previous versions to be more sensitive to tetracycline and its analogues.

Figures 1 and 2 below show expression from a promoter with six tetracycline response elements (TRE-Tight) in the presence of rtTA3 and various levels of doxycycline, a tetracycline analogue.
