Description

pToolkit is a key component of the alternative selection method (selection without antibiotics) proposed by iGEM2011 HKUST. The main function of pToolkit is to knock out one of the essential gene, nadE from E.coli's gDNA and replace it with part of the spilt sfGFP.

Notes

We construct the pToolkit by replacing its heat sensitive replication origin oriR101&RepA101 with pir gene regulated replication origin ori-gamma.

Source

The backbone of the pToolkit, including the lambda-red swapping system and the Ampicilin Resistance Gene comes from pKD46. The replication origin of the pToolkit is ori-gamma, from the relication origin R6K.
