Description
This biobrick contains the remaining 2 cellulose synthase enzyme complex, acsC and acsD of 4,363 bp in total (full CDS), again containing Xba I and Spe I restriction sites cloned in pSB1C3. The acsC gene is the main composite in the formation of the membrane complex of cellulose synthase and proposed to be involved in the export of the polymer across the bacterial cell wall.
The acsD gene is involved in the crystallization of the mature cellulose by cleaves the intrastrand ??-1,4 linkages in the cellulose chain and is proposed to have a role in the release of the growing polymer from the cell.
Notes
acsCD is long base pairs gene, so more than one primer is used during PCR. It is also advisable to separate PCR into a few times to get the most satisfied results.
