Description
This composite part constitutively expresses a ToxR-LambdaRep construct. This ToxR construct will dimerize at a high affinity, because of Lambda Rep's dimerizing properties, making the cytoplasmic ToxR dimer an active transcription factor for the ctx Promoter. Thus, If this composite part is combined with a Pctx-GFP reporter there will be a high expression of GFP.
Notes
The biggest design considerations were how to limit the amount of ToxR expressed while still allowing enough ToxR to be produced to see a high expression of GFP. The P_rffGH promoter, the rbs, and the plasmid we put it on (ColE1 origin) all contributed to the optimal expression level of the ToxR construct.
Source
The original promoter came from E. Coli genome, but this specific promoter was selected from a library of that promoter. This rbs was selected from a library in a tuning process. ToxR comes from a virulence island of Vibrio Cholerae. The Lambda Repressor is from the Lambda Phage.
