Description

We build this part to express the cI repressor, which can bind with the cI operator and accomplish more work.

We add Plac promoter on the 5' end. This promoter is consecutively expressed promoter.

Then, we add Lac operator between the promoter and cds, which will bind to lacI and inhibit the expression of cI protein. If you use this part in a lacI-coding plasmid like pet15b, the expression of gene will be repressed. However, IPTG can be used to activate the expression. The expression of this part can be well-controled.

This part can be used to construct a series of logic gate.

Notes

We want to control the expression of cI gene and construct some logic gate using this part.

We clone cI gene from other parts and add lac operator and Plac promoter by PCR step by step.
