Description
This shuttle vector is for use in the budding yeast Saccharomyces cerevisiae and the bacterium E. coli. This vector contains the f1 multiplecopy origin of replication for E. Coli and the CEN6/ARSH4 yeast origin of replication. For E. coli it has the ampicilline resistance and for yeast the auxotrophic selectable marker LEU2. The pRS415-II plasmid contains, in contrast to the previous pRS series, no illegal restriction site (EcorI, PstI, XbaI or SpeI) in the selectable marker site. The multiple cloning site (MCS) does contain these restriction sites, however the order of the restriction sites is different in comparison to the backbones that are compatible with the standard assembly method for biological parts (EPSX instead of EXSP).
Notes
Despite of the fact that this vector is not fully biobrick compatible it can be very usuful for the iGEM community.
