Description

Our research has focused onnovel biosynthetic pathways for UV-protectant compounds found in two distinct algal species. One leading to the production of the unrelated UV-screening compound scytonemin was cloned from Nostoc punctiforme. ScyA (NpR1276), an acetolactate synthetase, carries out the first step in scytonemin biosynthesis, converting L-tryptophan to alpha-keto-tryptophan.

This part includes a modified constitutive lac promoter (lacP'), and RBS and the open reading frame of scyA. This part can be used to express ScyA protein in E. coli.

Notes

N. punctiforme genomic DNA was obtained from ATCC. From here, PCR primer extension was used to clone the gene with flanking restriction digest sites to be used for vector cloning. The gene was cloned into an individual pUCBB BioBrick??? cloning vector containing a constitutive modified lac promoter (lacP???) and an RBS.
