Description
pCat promoter ([[BBa_I14033]]) flanked by regions ([[BBa_K137010]], [[BBa_K137008]]) recognized by fimE recombinase ([[BBa_K137007]]).
pCat is a constitutive promoter, which enables this device to be used as a switch to constitutively express two different regions. By default, the upstream sense strand is promoted. When flipped, the downstream antisense strand is promoted.
Additionally, downstream of the promoter is a terminating sequence [[BBa_B0015]]. This sequence is to prevent "backwards" transcription before the promoter is flipped. It is between the recombinant regions, causing it to flip with the promoter.
pCat can be replaced with another promoter for additional regulation if needed.
Notes
The double terminator placed before the promoter prevents any transient backwards transcription. Although the amount of transcription would be small, this design allows for stronger "digital" behavior.
