Description
This is a conjugative plasmid pSC101 with mobA (relaxase coding gene) and oriT (origin of transfer) knocked out, providing all other necessary proteins for conjugative transfer of plasmid pSC101 in the help of conjugative plasmid R751. The plasmid is a mutant, unable to perform conjugative functions, but when pSC101 MobA protein is supplied in the R751 strain, any plasmid with pSC101 oriT transfer origin in this chasis can be conjugatively transfered into recipient cells.
Notes
Region of deletion: 2936bp - 4115bp (containing mobA and oriT) and substituted with PstI restriction site CTGCAG
Method of deletion: in vitro reversed cyclic PCR and self-ligation.
