Description
This part is a reporter containing the lambda promoter Pr from the lambda switch flanked by a fluorescent reporter gene on each side. DsRED is on the side of Pr promoted by the protein cI, and acGFP is on the side promoted by Cro. An E. coli cell containing this plasmid and infected with lambda phage will fluoresce green if the lytic pathway is chosen or red if the cell becomes lysogenic.
Notes
During design we had to account for differing strengths of RBS in either fluorescent protein gene, since we wanted both genes to be expressed equally. We also had to modify the bistable switch from lambda so that it was less cumbersome.
Source
AcGFP and dsRED are commercial genes with commercial sequences. The lambda sequence was obtained from literature.
